چکیده
Synthetic ∆ 9 -tetrahydrocannabinol (THC), pharmaceutical grade herbal cannabis as well as formulations of well-defined cannabis extracts are available as registered medicines in several countries. It is generally accepted that the main central and peripheral pharmacological effects of THC are mediated by CB1 and CB2 cannabinoid receptors. Cytochrome P450-mediated oxidations of THC afford the non-psychoactive 11-nor-9-carboxy-THC (THC-COOH) as main metabolite. The pharmacokinetics of THC-COOH has been studied thoroughly and this abundant acid metabolite has become an established urinary marker of cannabis consumption in forensic, clinical and environmental analyses. Surprisingly, however, data on the biological activity of this ubiquitous metabolite are scarce. A few studies have examined the effect of THC-COOH on the biosynthesis of prostaglandins and other eicosanoids, on capsaicin-sensitive sensory nerves, on the multidrug transporter P-glycoprotein, on the cannabinoid and estrogen receptors in vitro, as well as its anticataleptic, analgesic, platelet-activating factor inhibitory and anti-inflammatory activities in vivo; THC-COOH has also been reported to block certain behavioural effects of THC in rodents. Hereby we provide a literature review on the reported pharmacological effects of THC-COOH and advocate further studies to reveal any potential involvement of this abundant metabolite in the complex pharmacology and in the proven therapeutic effects of THC-containing preparations.

Estimating the time of last cannabis use from plasma delta9-tetrahydrocannabinol and 11-nor-9-carboxy-delta9-tetrahydrocannabinol concentrations.
BACKGROUND Knowing the time cannabis was last used is important for determining impairment in accident investigations and clinical evaluations. Two models for predicting time of last cannabis use from single plasma cannabinoid concentrations-model I, using Delta(9)-tetrahydrocannabinol (THC), and model II, using the concentration ratio of 11-nor-9-carboxy-THC (THCCOOH) to THC-were developed and...

To drug-free urine specimens, we added the following drugs of abuse to give concentrations twice the cutoff value for positive test results: 11-nor-9-carboxy-delta-9-tetrahydrocannabinol (9-carboxy-THC), oxazepam, secobarbital, morphine, benzoylecgonine, amphetamine, or phencyclidine (PCP). Visine was then added. Although measured concentrations of several drugs were decreased in the presence o...

(-)-∆9-Tetrahydrocannabinol is the principal psychoactive component of the cannabis plant and also the active ingredient in some prescribed drugs. To detect and control misuse and monitor administration in clinical settings, reference samples of the native drugs and their metabolites are needed. The accuracy of liquid chromatography/mass spectrometric quantification of drugs in biological sampl...
متن کاملCan Physical Exercise or Food Deprivation Cause Release of Fat-Stored Cannabinoids?
The aim of this study was to evaluate whether physical exercise or food deprivation may increase cannabinoid levels in serum or urine in abstinent chronic cannabis users. The study took place in a drug detoxification ward parallel to study participants receiving treatment. Six chronic, daily cannabis users (one female, five males, average age 30.0 years; BMI 20.8) were exposed to a 45-min. mode...
متن کاملPhase I and II cannabinoid disposition in blood and plasma of occasional and frequent smokers following controlled smoked cannabis.
BACKGROUND Δ(9)-Tetrahydrocannabinol (THC), 11-hydroxy-THC (11-OH-THC), and 11-nor-9-carboxy-THC (THCCOOH) have been reported in blood from frequent cannabis smokers for an extended time during abstinence. We compared THC, 11-OH-THC, THCCOOH, cannabidiol, cannabinol, THC-glucuronide, and 11-nor-9-carboxy-THC-glucuronide (THCCOO-glucuronide) blood and plasma disposition in frequent and occasiona...
